    Design and implementation of a multi-modal biometric system for company access control

    This paper is about the design, implementation, and deployment of a multi-modal biometric system to grant access to a company structure and to internal zones in the company itself. Face and iris have been chosen as biometric traits. Face is feasible for non-intrusive checking with a minimum cooperation from the subject, while iris supports very accurate recognition procedure at a higher grade of invasivity. The recognition of the face trait is based on the Local Binary Patterns histograms, and the Daughman\u2019s method is implemented for the analysis of the iris data. The recognition process may require either the acquisition of the user\u2019s face only or the serial acquisition of both the user\u2019s face and iris, depending on the confidence level of the decision with respect to the set of security levels and requirements, stated in a formal way in the Service Level Agreement at a negotiation phase. The quality of the decision depends on the setting of proper different thresholds in the decision modules for the two biometric traits. Any time the quality of the decision is not good enough, the system activates proper rules, which ask for new acquisitions (and decisions), possibly with different threshold values, resulting in a system not with a fixed and predefined behaviour, but one which complies with the actual acquisition context. Rules are formalized as deduction rules and grouped together to represent \u201cresponse behaviors\u201d according to the previous analysis. Therefore, there are different possible working flows, since the actual response of the recognition process depends on the output of the decision making modules that compose the system. Finally, the deployment phase is described, together with the results from the testing, based on the AT&T Face Database and the UBIRIS database

    Relationship between Protein Oxidation Biomarkers and Uterine Health in Dairy Cows during the Postpartum Period

    High neutrophil (PMN, Polymorphonuclear neutrophil) counts in the endometrium of cows affected by endometritis, suggests the involvement of oxidative stress (OS) among the causes of impaired fertility. Protein oxidation, in particular, advanced oxidation protein products (AOPP), are OS biomarkers linked to PMN activity. To test this hypothesis, the relationship between protein oxidation and uterus health was studied in thirty-eight dairy cows during the puerperium. The animals were found to be cycling, without any signs of disease and pharmacological treatments. PMN count was performed either through a cytobrush or a uterine horn lavage (UHL). Cows were classified into four groups, based on the uterine ultrasonographic characteristics and the PMN percentage in the uterine horns with a higher percentage of high neutrophil horn (HNH). They were classified as: Healthy (H); Subclinical Endometritis (SCE); Grade 1 Endometritis (EM1); and Grade 2 Endometritis (EM2). AOPP and carbonyls were measured in plasma and UHL. UHL samples underwent Western blot analysis to visualize the carbonyl and dityrosine formation. Plasma AOPP were higher (p < 0.05) in EM2. AOPP and carbonyl group concentrations were higher in the HNH samples (p < 0.05). Protein concentration in the UHL was higher in the EM2 (p < 0.05). Carbonyl and dityrosine formation was more intense in EM1 and EM2. Protein oxidation observed in the EM2 suggests the presence of an inflammatory status in the uterus which, if not adequately hindered, could result in low fertility

    Subacute Ruminal Acidosis and Evaluation of Blood Gas Analysis in Dairy Cow

    Subacute Ruminal Acidosis (SARA) corresponds to an imbalance between lactate-producing bacteria and lactate-using bacteria, which results in a change in ruminal pH associated with a prevalent consumption of rapidly fermentable carbohydrates. In our study, 216 primiparus and multiparus dairy cows were selected from 20 Italian intensive dairy herds and were divided into three groups based on the risk of SARA. All the dairy cows had high average milk production. After blood sampling, a complete blood gas analysis was performed. One-way ANOVA was performed to compare the three groups. O2 Cont, PCO2, blood pH, O2Hb, urinary pH, and rumen pH were significantly lower in cows with rumen pH < 5.5. These results indicate that blood gas analysis is a valuable tool to diagnose acidosis in dairy cows because it provides good assessment of acidosis while being less invasive than rumen pH analysis

    Mild exposure of RIN-5F &#946;-cells to human islet amyloid polypeptide aggregates upregulates antioxidant enzymes via NADPH oxidase-RAGE: An hormetic stimulus

    AbstractThe presence of amyloid aggregates of human islet amyloid polypeptide (hIAPP), a hallmark of type 2 diabetes, contributes to pancreatic β-cell impairment, where oxidative stress plays a key role. A contribution of NADPH oxidase to reactive oxygen species (ROS) generation after cell exposure to micromolar concentrations of hIAPP aggregates has been suggested. However, little is known about β-cells exposure to lower amounts of hIAPP aggregates, similar to those found in human pancreas. Thus, we aimed to investigate the events resulting from RIN-5F cells exposure to nanomolar concentrations of toxic hIAPP aggregates. We found an early and transient rise of NADPH oxidase activity resulting from increased Nox1 expression following the engagement of receptor for advanced glycation end-products (RAGE) by hIAPP aggregates. Unexpectedly, NADPH oxidase activation was not accompanied by a significant ROS increase and the lipoperoxidation level was significantly reduced. Indeed, cell exposure to hIAPP aggregates affected the antioxidant defences, inducing a significant increase of the expression and activity of catalase and glutathione peroxidase. We conclude that exposure of pancreatic β-cells to nanomolar concentrations of hIAPP aggregates for a short time induces an hormetic response via the RAGE-Nox1 axis; the latter stimulates the enzymatic antioxidant defences that preserve the cells against oxidative stress damage

    The emerging role of the RBM20 and PTBP1 ribonucleoproteins in heart development and cardiovascular diseases

    Alternative splicing is a regulatory mechanism essential for cell differentiation and tissue organization. More than 90% of human genes are regulated by alternative splicing events, which participate in cell fate determination. The general mechanisms of splicing events are well known, whereas only recently have deep-sequencing, high throughput analyses and animal models provided novel information on the network of functionally coordinated, tissue-specific, alternatively spliced exons. Heart development and cardiac tissue differentiation require thoroughly regulated splicing events. The ribonucleoprotein RBM20 is a key regulator of the alternative splicing events required for functional and structural heart properties, such as the expression of TTN isoforms. Recently, the polypyrimidine tract-binding protein PTBP1 has been demonstrated to participate with RBM20 in regulating splicing events. In this review, we summarize the updated knowledge relative to RBM20 and PTBP1 structure and molecular function; their role in alternative splicing mechanisms involved in the heart development and function; RBM20 mutations associated with idiopathic dilated cardiovascular disease (DCM); and the consequences of RBM20-altered expression or dysfunction. Furthermore, we discuss the possible application of targeting RBM20 in new approaches in heart therapies

    Glucose infusion response on some metabolic parameters in dairy cows during transition period

    Abstract. The aim of this study was to evaluate the effect of glucose tolerance test (GTT) on glucose, insulin, non-esterified fatty acids (NEFA) and β-Hydroxybutyrate (BHB) concentration in dairy cows during prepartum and postpartum period. Forty Two clinically healthy Holstein cows were enrolled in two high producing dairy farms in the Northeast Italy. All animals were divided into two equal groups on the basis of the farm of origin: Group A (farm located in Vicenza) and Group B (farm located in Padua). Body condition score (BCS) was determined for both groups. GTT was performed 7 ± 5 days before and 7 ± 5 days after calving. Blood samples were collected from each animal before (T0) and after the administration of glucose at different times. Plasma concentration of glucose, insulin, NEFA and BHB was evaluated. Application of two way analysis of variance (ANOVA) showed no significant effect of farm and a significant effect of transition period (prepartum and postpartum) on BCS for both groups. Multivariate analysis of variance (MANOVA) was applied to determine significant effects of infusion glucose time, transition period and farm on glucose, insulin, NEFA and BHB. No significant difference was observed between Groups A and B and a significant effect of transi tion period and glucose infusion was found on all parameters studied. Results confirm that glucose is an important direct controller of metabolic response in dairy cows during the transition period because of the inappropriate energetic supply that characterizing this physiological phases

    Dissection of HLA-C gene region to investigate its association with complex traits

    The genomic region of HLA gene cluster (6p21.3) contains several highly polymorphic genes involved in the immune response and therefore, they have been previously associated to several immune and inflammatory disorders. For the present study we investigated more than 3500 known alleles of HLA-C gene (from IPD-IMGT/HLA database, genomic sequences) that are grouped into 14 serogroups (e.g., C*01:02:01:01). All the sequences have been aligned against the human genome reference sequence (both versions; hg19 and hg38). Overall, the HLA-C gene (length ~3000 bp) contains more than 1500 SNPs. We used a clustering approach to understand how the alleles are evolutionarily connected. According to the clustering analysis we observed that sequences of the same serogroup cluster together more often than sequences of other serogroups, even if with several exceptions. This confirms that alleles of the same serogroup share strong identity in their sequence. Interestingly, as general rule, we observed that the main tree presents two branches, containing each a similar structure (relative distance between serogroups). Of note, the sequences of C*07 and C*17 serogroups belonged to one of the two branches only, whereas the sequences of C*06 and C*12 serogroups were observed in the alternative branch of the tree. We are now studying what are the main features that characterize the two branches. Moreover, the study will go on by investigating the association of the HLA-C serogroup SNP-based alleles with kidney related disease (INCIPE study) and Alzheimer’s disease (NIAGADS database) in large cohorts of individuals

    Disease-specific and general health-related quality of life in newly diagnosed prostate cancer patients: The Pros-IT CNR study

    Background: The National Research Council (CNR) prostate cancer monitoring project in Italy (Pros-IT CNR) is an observational, prospective, ongoing, multicentre study aiming to monitor a sample of Italian males diagnosed as new cases of prostate cancer. The present study aims to present data on the quality of life at time prostate cancer is diagnosed. Methods: One thousand seven hundred five patients were enrolled. Quality of life is evaluated at the time cancer was diagnosed and at subsequent assessments via the Italian version of the University of California Los Angeles-Prostate Cancer Index (UCLA-PCI) and the Short Form Health Survey (SF-12). Results: At diagnosis, lower scores on the physical component of the SF-12 were associated to older ages, obesity and the presence of 3+ moderate/severe comorbidities. Lower scores on the mental component were associated to younger ages, the presence of 3+ moderate/severe comorbidities and a T-score higher than one. Urinary and bowel functions according to UCLA-PCI were generally good. Almost 5% of the sample reported using at least one safety pad daily to control urinary loss; less than 3% reported moderate/severe problems attributable to bowel functions, and sexual function was a moderate/severe problem for 26.7%. Diabetes, 3+ moderate/severe comorbidities, T2 or T3-T4 categories and a Gleason score of eight or more were significantly associated with lower sexual function scores at diagnosis. Conclusions: Data collected by the Pros-IT CNR study have clarified the baseline status of newly diagnosed prostate cancer patients. A comprehensive assessment of quality of life will allow to objectively evaluate outcomes of different profile of care
